Abstract

The characteristics of nanozeolite-derived enzyme microreactor using zeolite nanocrystals as substrates for protein immobilization are investigated. Two model proteins such as trypsin and cytochrome c, with different concentration and molecular weight were used to evaluate the efficiency of microfludic reactor. Zeolite LTL nanocrystals was used as trypsin-immobalizing carrier and was patterned on poly (methyl methacrylate)(PMMA) microfludic channel to facilitate immobilization of nanozeolite particles in the microchannel. The influence of immobilization capacity of nanozeolite LTL on the proteolytic efficiency of microreactor, adsorption capacity of nanozeolites were studied by TEM and MALDI-TOF MS. It was found that adsorption capacity of the nanozeolites for trypsin increases with increase in its concentration. Result shows that microreactor is capable of efficient protein digestion at low concentration in short reaction time and has good stability for protein identification.
